Many pharmaceuticals currently on the market are preparations for oral administration, many of which are stable in the gastrointestinal tract and have good absorption from small intestinal epithelial cells. Drug absorption from the gastrointestinal tract is the first step in the pharmacokinetics, and with the exception of some exceptions, no drug efficacy can be expected unless the drug is absorbed. However, in recent drug development, poorly absorbable drugs such as poorly water-soluble drugs and high molecular weight drugs are increasing, and various studies have been conducted to improve the absorption of these drugs in the gastrointestinal tract. These methods can be broadly divided into two. One is an absorption enhancer that acts on the gastrointestinal tract to promote permeation of substances. The other is to improve the physicochemical properties of the drug, such as water-solubility and fat-solubility, by chemically modifying the drug or devising the dosage form. In this symposium, we will talk about various efforts to improve gastrointestinal absorption of these drugs, and discuss future oral formulation.
