[S31] Latest findings of non-P450 drug-metabolizing enzymes aimed for drug discovery and clinical applications

Drug-metabolizing enzymes play a central role in pharmacokinetics by drug inactivation, but are sometimes involved in the onset of drug-induced toxicity by a production of reactive metabolites. Therefore, the function of the enzymes should be characterized from the aspect of both drug efficacy and toxicity. Among drug-metabolizing enzymes, cytochrome P450 enzymes have been well studied, because they metabolize many kinds of medicines used in clinical practice. In recent years, it has tended to avoid the development of drug candidates metabolized by P450 enzymes due to drug-drug interactions. Moreover, such compounds occasionally show unexpectedly high systemic clearance observed in clinical trials compared with those estimated by preclinical studies. From these points, non-P450 enzymes are attracting attention in drug discovery stages. However, there is still being elucidated about their function and regulatory factors. This symposium introduces novel findings about non-P450 enzymes from viewpoints of fundamental researches, and discuss how to apply non-P450 enzyme research in drug development and appropriate use of drugs in clinical practice.
