Multi-layered thin films with heterogeneous mechanical properties can be harnessed to transform into various three-dimensional (3D) geometries spontaneously. Here, we demonstrated micropatterned all-polymer films, termed micro-rolls, for encapsulating, manipulating, and observing adherent cells in vitro. The micro-rolls have double layered films consisting of two polymers possessing different mechanical stiffness; thereby they can be fabricated by the strain engineering and self-folding roll-up process. The geometrically controlled strain of nano-films achieves 3D tubular architectures with controllable diameter. Thus, the multiple cells can be wrapped in individual micro-rolls and artificially reconstructed into hollow or fibre-shaped cellular 3D constructs that possess intrinsic morphologies and functions of living tissues. This system can potentially provide 3D bio-interfaces such as the reconstruction and assembly of functional tissues and implantable tissue grafts.
