[15a-4E-6] Organelle distribution in SX image by correlation between SX and fluorescence images

Soft X-ray (SX) imaging using Water-window wavelength region (l=2.34 - 4.4 nm) can image bio-specimens in water solution and show high spatial resolution than in visible wavelength region. The fine structures photographed in a SX image show different contrast comparing with visible images because of different extinction coefficients. Therefore identification of the fine structures imaged in Water-wavelength region is difficult by its appearance only. In this study, a SX image of Leydig cells of a mouse testis is compared with fluorescence ones of the same cells at the same position. The cells were dyed by fluorophores to identify some organelle in the cells. Identification of fine structures in the SX image is carried out by comparison with fluorescence ones with the use of principal component analysis for images. The result shows that the common structures between the SX and fluorescence images dyed by Phalloidin coincide well with a part of the fine structures observed in the SX image.
