[15p-2A-5] Discrimination of three nucleotides in single stranded DNA with a solid state nanopore and Active Control Method

In developing nanopore DNA sequencing, controlling DNA translocation is an essential technique. We discriminated three nucleotides in single stranded DNA with using solid state nanopore and active control method. For active control method, we used DNA immobilized probe whose size is larger than membrane fabricated nanopore and adopted stepper motor of Scanning Tunneling Microscope for driving nanopore substrate. Using this technique user need not control precise relative position between nanopore substrate and probe. In this presentation, we demonstrate that DNA (block copolymer; ([(dT)25-(dC)25-(dA)50]m)) immobilized on the probe slid through nanopore and is arbitrary pulled out from nanopore. During driving nanopore substrate, we obtain different current signal level corresponding to each kind of nucleotide.
