[14p-P23-10] Enhanced crystallization of proteins induced by laser trapping at the air/water interface

Focusing of a 1064-nm laser beam to an air/solution interface of a lysozyme solution induces assembly formation, many micrometer-sized dense liquid droplet formation due to a liquid-liquid phase separation and, eventual crystallization after droplet formation. Raman microspectroscopy under trapping laser irradiation suggests that the supersaturation degree of the solution ascends rapidly due to the local concentration elevation by trapping of lysozyme clusters. However, real-time crystallization of protein from the focal spot by laser trapping crystallization method has not been realized. We observed real-time crystallization of cage shaped supramolecular protein, ferritin. We will discuss different crystallization behavior between lysozyme and ferritin by means of Raman micro spectroscopy.
