We previously developed a multipoint holographic fluorescence correlation spectroscopy (MP-hFCS). In the system, a problem was expected that the phototoxicity induced by multipoint excitation laser would increase with increasing of the measurement points of MP-FCS. To overcome this problem, a MP-FCS using two photon excitation (multiphoton MP-FCS, MP-hFCS) was developed in this work. With the two photon excitation, it can be expected that the phototoxicity is suppressed to least level because excitation laser is absorbed only within the focal region. In this report, we discussed the measurement results of fluorescence dye solution and the measurement inside plants.
