In our group, thin-metal coated grating substrate, i.e., plasmonic chip, has been used for the grating-coupled surface plasmon resonance-field enhanced fluorescence and it has been studied for sensitive bio-detection and fluorescence bio imaging. In our previous presentation, the Bull's eye plasmonic chip was shown for more useful structure under the fluorescence microscopic observation. In this study, for the observation of a single nano-particle, the fluorescence enhancement factor on the plasmonic chip was improved by the control of the illumination angle at aperture stop of a microscope.
