[17p-413-4] In-situ infrared micro-spectroscopic study of metabolic mechanism on living cells in medium solution.

In-situ transmission infrared (IR) absorption measurements were performed for living cell lines of mouse (NIH3T3 and B16). Incident infrared ray attenuates rapidly in water using traditional transmission method, then, the transmission IR micro-spectroscopy in aqueous condition is not common. We fabricated novel optical cell which has 12 μm-thick medium solution layer and cell lines in optical path. Therefore, we succeeded to obtain transmission IR spectrum from living cells without influence of absorption of water molecules. According to the analysis of IR absorption spectrum from two kinds of living cells, we found two significant differences due to the metabolic mechanism, the amount of lactic acid and adenosine triphosphate (ATP). Therefore, we suggest that this method can be applied to detect a metabolic disorder in cell level.
