This study investigated the effect of three kinds of underlying substrates on the artificial supported lipid membrane (SLB) with using atomic force microscopy (AFM) and a diagonal illumination setup for single particle tracking (SPT). SLB is an artificial cell membrane system, in which physical properties of lipid bilayer are key factors affecting the functions and activities of membrane proteins. The detailed observation at single molecule level revealed that the membrane surface structures and the diffusion of incorporated dye lipids depend on the kinds of substrate. The consideration of the interactions between the substrate and SLB based on our results is important for the construction of SLB systems.
