Acetaldehyde-derived nucleic acid base N2-Et-dG formed by the addition of ethanol metabolite to deoxyguanosine (dG) has attracted attention as a marker for esophageal cancer, However, complicated p preprocessing is necessary for detection of N2-Et-dG. In this study, single-molecule measurement of N2-Et-dG were performed with the aim of developing a preprocessing-free N2-Et-dG detection method.
In the experiment, the modified nucleic base N2-Et-dG was detected using single-molecule measurement. We succeeded in discrimination between N2-Et-dG and dG single-molecule signals by using of machine learning. The possibility of single molecule measurement as a detection method of N2-Et-dG was presented.
