[19p-PB9-11] Microfabricated SiN-based solution chamber for Diffracted X-ray Tracking method

Diffracted X-ray Tracking (DXT) method enables us to track dynamic conformational change of ion channel by irradiating gold nanocrystal-attached ion channel with white X-ray and tracking motion of diffraction spot from gold nanocrystal. In order to decrease background noise which prevented observation of diffraction spot in previous DXT setup, we developed novel solution chamber with microfabricated silicon nitride membrane as observation window. DXT measurement of pH-dependent ion channel (KcsA) was conducted under the different pH conditions with this chamber. Trajectory of diffraction spot was clearly observed, enabling to analyze dynamic gating motion effectively.
