[10a-M116-2] Subtraction Imaging using Higher-Order Vector Beams for Biological Samples

We develop a new method that can improve the spatial resolution in confocal laser scanning microscopy (CLSM) simply by subtracting two images acquired by bright- and dark-spot-shaped point spread functions. We have previously demonstrated that the usage of higher-order vector beams such as radially and azimuthally polarized beams in subtraction imaging can dramatically improve the spatial resolution in CLSM with reduced negative side-lobe generation. In this study, we apply our method to biological samples and evaluate its imaging capability in terms of the impact of negative side-lobe generation.
