[10a-M116-3] Microspectroscopy of Diatom Frustule Using Micro-glassfiber Probe

An optical system of transmission mode microscopic spectroscopy was constructed using a pair of optical fibers sharpened to the micrometer scale as the microscopic light guide. Absorption spectra attributed to chlorophyll of chloroplasts of a diatom were measured with spatial resolution of about 20 μm by sandwiching a diatom cell between the light source side and the detecting side optical fiber ends. In addition, since the light flux is almost parallel in the fiber gap, we successfully evaluated the photonic crystal characteristics of a diatom frustule with anisotropy in propagation direction of light.
