[9a-W242-5] Epigenetic Discrimination of Nucleotide For Single-Molecule Tunnel-Current Electrical Identification

We developed a single-molecule identification method by using tunnel current measurement in order to discriminate chemically modified nucleobases for natural nucleobases. We found that obtained signals of tri-fluorouracil monomer were statistically discriminated from the other nucleobases based on the characteristic conductance values. In addition, oligonucleotide containing the trifluorauracil was measured by this method and its signals were assembled into the reference sequence. The results of conductance plots suggests that differences in sequence including trifluorouracil are discriminated from the obtained oligonucleotide without trifluorauracil molecules.
