[9p-W242-4] Rapid and sensitive detection of Candida mannan using the plasmonic sensor chip
Keywords:plasmonic chip, candida mannan, immunity
Oral candida contained in saliva has been indicated to be a marker for measuring immunity and . Currently, the number of oral candida bacteria is measured by ELISA and method or bacterial culture methods. However, these methods lack ofIn order to meet requirement of the quickness rapid and conveniencesensitive detection,. iIn this study, a sandwich assay was constructed on a plasmonic chip and measured candida mannan was quantitatively measured by grating-coupled surface plasmon-field enhanced fluorescence (GC-SPF).
The plasmonic chip was prepared by UV nano-imprint methoding and sputteringmetal coating. As a sandwich assay, aAnti-candida mnannman IgG antibody was immobilized toon the chip surface coated with protein G and polydopamine filmcoated chip with Protein G bound. After injections ofThe candida mannan in positive and negative control solutions was quantitatively measured with a fluorescent labeled antibodypositive or negative control solutions, sandwich assays were constructed using fluorescent labeled antibodies. Reflectance was measured for determining the SPR angle and fluorescence intensity at the SPR angle was measured. Coefficient of variation inof the measured fluorescence intensity was less than 0.10 or less. and These result showed that tthe mannan detection with a plasmonic sensor chip showed could be performed positive detection withthe high reproducibility.
