[10p-Z28-1] Single-cell imaging with mid-infrared photothermal quantitative phase imaging

Label-free optical imaging is suitable for long-time observation of live and unperturbed biological cells. Especially, molecular vibrational imaging and quantitative phase imaging, which offer molecular-structural chemical information and mass-density-based morphological information, respectively, have undergone intensive technological development in the past twenty years. Recently, we have developed a novel label-free imaging method called mid-infrared photothermal quantitative phase imaging, in which the two label-free methods are robustly integrated to circumvent their complementary limitations. In this contribution, we demonstrate the capability of our method to perform three-dimensional single-cell imaging in the broadband mid-infrared vibrational fingerprint region.
