In this study, the spatial resolution of a laser scanning transmission microscopy is improved by using saturated absorption of dyes. This phenomenon occurs locally near the center of the illumination spot and generates transmitted signal component that exhibits a non-linear response to the excitation light intensity. By performing imaging while selectively detecting non-linear transmitted signal response, it is possible to compose an image from a transmitted signal generated from a region smaller than the illumination spot. Images of stained tissue sections show that this method improves the spatial resolution of transmission microscopy.
