In this study, the label-free detection of methylcytosine using nanoimprint lithography-based two-dimensional photonic crystal was succeeded. For detection of methylcytosine, the probe DNA was immobilized onto PhC surface covalently. And the target DNA which contained methylcytosine was introduced. After the hybridization of probe DNA and target DNA, the optical characteristics change was monitored. As a result, the differences of optical characteristics between methylcytosine contained target DNA and methylcytosine without contained target DNA could be observed.
