It has been known that the density of environmental DNA of fish increases in the breeding season. On the basis of these empirical knowledge, we validated the eDNA analysis as a method to grasp a laying eggs activity of crucian carps in the reproduction reed zone maintained in the river mouth of the Yasu River in Shiga Prefecture. We conducted qPCR analysis for water samples which collected near a reproduction reed zone in the breeding season. Our results clearly demonstrated that the density of environment DNA of crucian carp became higher in the breeding season.
