[2Bp03] Development of AFM Tip with Fluorescent Diamond Nanoparticle for Intracellular 3D Temperature Mapping
Recent progress in 3D-AFM enables the nanoscale 3D imaging of living cell internal structures. However, the information obtained by 3D-AFM is limited to force distribution, and hence it is difficult to simultaneously investigate physical information other than mechanical properties. One possible solution to this problem is using a quantum sensor. In particular, the frequency shift of optically detected magnetic resonance (ODMR) of fluorescent nanodiamonds (NDs) is sensitive to ambient temperature. To combine it with 3D-AFM, here we developed techniques for measuring the ODMR of a fluorescent ND on an AFM tip. First, we established the method for fixing fluorescent ND on the AFM tip apex using a carbon gas injection system. Then, we implemented the custom-built 3D-AFM system, single photon counting module, and sample holder on the commercially available confocal microscope. With this system, we have succeeded in detecting the ODMR of fluorescent NDs scattered on the glass substrate.
