[S22] "Unconventional" drug-discovery research using platinum group metals

Even now, more than 40 years after the clinical introduction of cisplatin, platinum-based drugs are still used in chemotherapy for various solid tumors. All platinum-based drugs are electrically neutral cis-type mononuclear platinum(II) complexes, and if the molecular design is performed using the same basic structure, a platinum complex exhibiting an effective antitumor efficacy can be obtained. However, in recent years, with the aim of reducing side effects and overcoming cross resistance to platinum-based drugs, platinum complexes with different basic structure from platinum-based drugs, or platinum group elements other than platinum (ruthenium, rhodium, palladium, osmium, iridium) is being used for "unconventional" drug-discovery research. Since a metal complex of a platinum group element is not easily hydrolyzed, it can bind to a target biomolecule while maintaining a unique geometric structure peculiar to the metal complex in an aqueous solution. In addition, the introduction of a functional ligand enables drug discovery using an approach different from that of conventional platinum-based drugs. In this symposium, we will introduce the drug-discovery researches of cancer therapeutic agents and cancer diagnostic agents using platinum group elements, and discuss new developments in inorganic medicinal chemistry.
