[P3-203] Correlation between Receptor-interacting Protein 140 (RIP140) Expression and Neural Directed Differentiation from Human Embryonic Stem Cells to Neural Stem Cells

[Objective] To determine the expression profile of receptor interacting protein 140 (RIP140) during the directed differentiation from human embryonic stem cells (hESCs) to neural stem cells (NSCs) and analyze their correlationship. [Methods] Embryonic stem cells H1 was used to do the neural directed differentiation in vitro. Then the cells were observed by inverted phase contrast microscope, immunostained with special hESCs and NSCs markers on day0, 3.5, 7 respectively after induced with special differentiation media. Meanwhile, RIP140 expression was detected with immunofluorescence and real-time polymerase chain reaction (PCR).In combination with the expression of ERK1/2 protein, we analyze the correlation between RIP140/ERK and neural directed differentiation using one-way ANOVA. [Results] After induce differentiation, hESCs stretch many branches confusing eventually and present a neural stem cell-like characteristics. The hESCs marker Oct4, Sox2 decrease and NSCs marker Nestin increases during differentiation. RIP140 and ERK1/2 are also increased. RIP140 has a negative correlation with Oct4, Sox2 and a positive correlation with Nestin in late stage of differentiation. [Conclusion] RIP140 and ERK1/2 might be involved in the progress of neural directed differentiation from hESCs to NSC.
