Quantitative phase imaging (QPI) allows us to measure the complex electromagnetic wavefront and is therefore useful for label-free volumetric imaging, morphological imaging of transparent specimens, etc. However, QPI lacks chemical contrast which limits its applications to morphology-based diagnosis. In this report, we propose and demonstrate that mid-infrared photothermal effect can be utilized to obtain molecular vibrational information of the specimen in the framework of QPI. We demonstrate wide-field, label-free chemical imaging capability of our QPI system in the molecular vibrational fingerprint region of 1,450 – 1,640 cm-1 based on silica-polystyrene microbeads mixture and biological cells such as cos7.
