In this paper, we discuss the lateral diffusion of lipids and phase separation between liquid-ordered (Lo) and liquid-crystalline (La) phase separation at the freestanding lipid bilayer suspended over microwells. We found that the lateral diffusion through the boundary between the freestanding part and the supported part on the substrate is anisotropic. It is also indicated the existence of the large diffusion barrier at the boundary. These play an important role in the formation and retention of phase separation at the freestanding membrane.
