[9a-W641-11] High-speed, high-precision single molecule imaging of biomolecular motor with gold nanoparticles

金ナノ粒子をプローブに用いた、高速・高精度な生体１分子イメージング法の開発を行った。輪帯照明型の全反射暗視野顕微鏡を開発し、40nmの金ナノ粒子を観察した。1ミリ秒、及び33マイクロ秒の時間分解能で、それぞれ1.3オングストローム、5.4オングストロームの位置決定精度を達成した。開発した装置を用い、モータータンパク質キネシンのモータードメインの挙動を10マイクロ秒の時間分解能で追跡した。
