In the modern age where new drug modalities diversify from low-molecular-weight compounds to proteins, nucleic acids and cells, targeting ability to therapeutic sites is one of the most important additional functions for their successful clinical application. Recently, the approval of new drugs with physiological targeting ability based on the specific affinity of antibodies or ligands to target sites as well as the enhanced vascular permeability of macromolecules or nanoparticles in tumor or inflamed tissues has been steadily increasing. However, their targeting efficiencies can be decreased or varied by several factors including their distribution to undesired sites through unspecific interaction with biocomponents, the state in patients, and the progression degree of diseases. In contrast, physical drug targeting through local administration and stimulation is expected to more reliably achieve enhanced delivery or specific action of drugs in target sites, although comprehensive technical innovation in relevant formulations, delivery devices, and stimulation systems is essential for its further progression and wide acceptance. In the present symposium, we would like to show the newest technological outcomes for physical drug targeting through local administration and stimulation, followed by discussion with audiences about future targeting strategy.
