[S50] JAK-STAT Cytokine Signaling Pathway: From Discovery to Clinical Application

Cytokines are important humoral proteins that trigger a variety of biological responses in homeostatic and disease states. Their signaling is mediated by the JAK-STAT pathway, which involves four Janus kinase (JAK) proteins downstream of their receptors and seven Signal Transducer and Activator of Transcription (STAT) proteins, and is deeply involved in diverse aspects of innate immunity, adaptive immunity, inflammation, stem cells, developmental biology, and cancer biology. In the 30 years since their discovery, our understanding of how these signaling pathways are regulated, function, and integrate with other cellular responses has deepened and refined. Drug discovery efforts have yielded small molecules targeting JAKs and STATs, which are now finding clinical application in the treatment of chronic skin diseases, autoimmune diseases, tumors, and many other inflammatory diseases, as well as in the treatment of severe COVID-19. In this symposium, researchers at the forefront of JAK-STAT research will present their research efforts and results, as well as novel findings from clinical practice, to provide further perspectives and challenges for therapeutics targeting JAK-STAT signaling.
