[P3-32] Analysis of cytokines in cerebrospinal fluids in enteroviral meningitis with or without pleocytosis in children

[Introduction] To compare the levels of cytokine profiles in cerebrospinal fluid (CSF) from children with enteroviral meningitis (EVM) between CSF pleocytosis and non-pleocytosis. [Methodology] Enterovirus was examined by real-time PCR in CSF for the children who underwent a lumbar puncture for suspected meningitis between 2015 and 2016. Total 131 patients were classified to the EVM with pleocytosis (group-A, n=71), EVM with non-pleocytosis (group-B, n=30), and non-meningitis (control, group-C, n=30) groups. Fifteen cytokines/chemokines, namely interleukin (IL)-1β, IL-2, IL-4, IL-5, IL-6, IL-7, IL-8, IL-10, IL-12, IL-13, IL-17, interferon (IFN)-γ, tumor necrosis factor (TNF)-α, granulocyte colony-stimulating factor (G-CSF), granulocyte monocyte colony-stimulating factor (GM-CSF), were measured simultaneously in CSF by immunoassay. [Results] The mean age was significantly younger in children of group-B (3.4±2.8 years) than in them of group-A (5.5±3.2 years, p=0.012). The levels of IL-1β, IL-2, IL-4, IL-6, IL-8, IL-10, IFN-γ, and TNF-α were significantly increased in EVM patients regardless of pleocytosis as compared to group-C. The levels of IL-1β, IL-2, IL-6, IL-8, IL-10, IFN-γ and TNF-α were significantly higher in group-A than in group-B. Especially, the levels of IL-2, IL-6, IL-8, and TNF-α were the highest in group-A, followed by group-B and group-C. Of all cytokines examined, CSF levels of IL-2, IL-6, IL-10 and TNF-α were correlated with pleocytosis in the EVM. [Conclusions] Significant elevation of CSF cytokine in EVM can be markers of meningeal inflammation when compared with control group. The significant difference between CSF pleocytosis and non-pleocytosis in EVM can be correlated with distinct levels of CSF cytokines.
