[V-16-20] Evaluation of anti-inflammatory immunobiotics in bovine mammary epithelial cells

[Objective] Bovine mastitis, also known as the inflammation of the mammary gland, is one of the most economically devastating diseases in dairy cattle. In this study, we focused on immunobiotics to prevent mastitis and aimed to evaluate useful immunobiotics against mastitis. [Methods] We evaluated the anti-inflammatory effects of 50 Lactobacillus strains to select the immunobiotics candidates. Bovine mammary epithelial (BME) cells were stimulated with the selected strains, followed by LPS, and then analyzed for immune-related gene and TLRs signaling factors using RT-qPCR and Western blotting. In addition, we evaluated the effects of the selected strain on the expression of milk protein genes in differentiated BME cells. [Results] Stimulation of a selected strain before LPS stimulation to BME cells down-regulated the expression of several cytokines and inhibited IκBα degradation and p38 pathway. In addition, the influence of LPS on milk protein gene expression was suppressed by pre-stimulation of a selected strain. These results show great promise for the developmental use of immunobiotics for the prevention of mastitis especially caused by E. coli.
