[V-20-06] Population structure in Japanese wild boar (Sus scrofa leucomystax) by maternal and paternal lineage genetics

It is interesting to analyze the unique genetic differentiation and adaptive evolution of Japanese wild Boar, as a geographically isolated subspecies. Thus, it can help to understand evolution of the genus Sus in Asia and highlight it in line with wildlife control measures. Therefore, we investigated genetic structure of JWB and phylogenetic relationships with other WB populations by analyzing mtDNA D-Loop and Y-chromosome from 96 JWB samples from Honshu, Shikoku and Kyushu. Consequently, we identified 22 Asian mtDNA haplotypes, although 1 animal phenotypically identified as wild boar had European domesticated pig haplotype. In Y-chromosome, 22 SNPs and 3 indels investigated were monomorphic whereas, we identified 2 novel Y-SNPs and 3 Y-indels in JWB. Interestingly, the JWB showed one of SRY haplotypes that may represent the putative ancestral haplotype, as observed in the aligned SRY sequences from Suidae Family. In contrast, other Y-SNPs seems to agree with an expansion of a non-Asian Y-haplogroup in JWB. In conclusion, JWB showed Asian mtDNA haplotypes and higher variability in contrast to Y-chromosome, where 2 novel Y-SNPs and 3 Y-indels were found.
