[3Ca04] Elucidation of pathological features of Parkinson’s disease using human induced pluripotent stem cell-derived region-specific neurons

The brain is a well-organized organ consisting of various brain regions. Since human pluripotent stem cells (hPSCs) have an ability to generate all cell types, region-specific neuronal differentiation from hPSCs would help for the disease modeling. During the last years, we investigated cell type composition and profiles of the developing human ventral midbrain by single cell RNA-sequencing. We subsequently established an induction protocol for midbrain dopaminergic (mDA) neurons from hPSCs by resembling the crucial factors we found from the dataset. Moreover, we also established a protocol for generation of striatal GABAergic neurons from hiPSCs. Next, we developed striatal and midbrain neurospheres from hPSCs using three-dimensional culture system. We observed the specific neuronal subtypes that are presented in each brain region. These region-specific neurospheres enable to investigate for the elucidation of the pathological features of the patients with Parkinson’s disease.
