In the development of chemical substances such as pharmaceuticals, safety assessment using laboratory animals are generally required and the results are often emphasized. On the other hand, several Western countries have banned animal testing in the development of cosmetics based on the perspective of the animal welfare 3Rs (Replacement, Reduction, Refinement). In recent years, Japan has also been considering a move to ban animal examinations not only in cosmetics but also in food additives. Due to this background, the development and practical application of alternative methods to animal examinations are needed. However, all the in vitro safety assessment tests generally used so far are 2D-systems using single cells, which have limitations in terms of extrapolation to humans. The development of a Micro physiological System (MPS) using 3D culture techniques is attracting attention as an innovation to break through this limitation. In this symposium, based on the viewpoint of regulatory science, we will have 6 speakers involved in the development and safety evaluation of chemical substances such as pharmaceuticals using tissue- and iPS cell-derived organoids/spheroids, and they will talk on the status and prospects of MPS development using 3D culture methods.
