Various molecular networks are activated in diseases. In this symposium, we will focus on dynamism in the activation state of molecular networks in diseases caused by changes in molecular expression levels, activities, and interactions. Recent advances in development of Adverse Outcome Pathways (AOP) and New Approach Methodologies (NAMs) in the OECD etc., will be explained. Introducing the subsequent development of the symposium S52 "Molecular Pathway-based Prediction of Adverse Events in OECD etc." at the 139th Annual Meeting of the Pharmaceutical Society of Japan held in 2019, the status of international joint research on AOP development brought about by molecular network research, its application in various fields will be summarized. In addition, we will introduce the perspectives and status of immunotoxicity-related initiatives in AOP development and OECD testing guidelines and discuss database analysis and machine learning analysis of AOP elements such as Molecular Initiating Event (MIE) and Key Event (KE) related to adverse drug reactions and drug activity prediction for drug discovery. We will discuss NAMs, which have been expanding further in recent years, including AOP networks and quantitative structure activity relationships (QSAR) as main topics.
